Therapeutic Effects of Cyclophosphamide in the Experimental Silicosis.


Abstract
In order to study of cyclophosphamide response in the experimental silicosis, 120 male rats weighed about 180gm were divided into six groups which a group contained 20 rats. To all of groups, free silica dust 50 §· solved in 0.5§¢ of physiological saline solution was given intratracheally and these subjects have observed 150 dayslong.
The control group wich was given only 50§· free silica solution was sacrificed after 150 days and their lung tissues was made pathlogical slides with Hematoxilin Eosin stain. The experimental group 1 was daily given 5§· cyclophosphamide intramuscularly seven times from 151th day after administration of free silica solution, and two weeks after injection, the experimental animals were sacrificed.
The experimental methode of group 2 was same to group 1 except four weeks instead of two weeks in group 1 after cyclophosphamide injection. The experimemtal group 3 was handled similar to group 1, as well as dosage of cyclophosphamide and the sacrificed time after its injection, but to this group, Cyclophosphamide as in primary was injected secondary after a week. The experimental methodes of group 4 and 5 were almost similar to group 1 in former and to group 3 in latter, except additional injection of 1§· steroid at each time of cyclophosphamide to be given.
The results were as follows;
1. The silicotic granulomas occured in all cases of the control group.
2. The severity of silicotic granulomas in only cyclophosphamide given group were milder along to the observed duration after cyclophosphamide and its total dosage.
3. The silicotic granulomas were very mild or not occured in the groups wich cyclophorphamide and steroid were additionally given.
4. In the groups to be given steroide, gangrene and/or lung abscess were occured more frequently than in the group without steroid.
